Monoclonal Anti-b-Tubulin, Isotype III, Clone SDL-3D10 (T 8660)

Whole cell extract of rat brain cytosol was separated on SDS-PAGE and blotted with Monoclonal Anti-b-Tubulin
Isotype III (Product No. T 8660). The antibody was developed with Goat anti-Mouse IgG, Peroxidase conjugate (Product No. A 9917) and AEC substrate.

Lane A: Antibody dilution 1:400

Lane B: Negative control (only secondary antibody)
Enhanced Validation-By Genetic Strategies HeLa TUBB3 KO (knocked out) Cell Lysate vs. HeLa wild type Cell Lysate were separated on SDS-PAGE and probed with 1:1,000 Monoclonal Anti-β-Tubulin III antibody produced in Mouse (Cat. No. T8660). Anti-GAPDH antibody, Mouse monoclonal, clone: GAPDH-71.1 (Cat. No. G8795) was used as loading control. The antibodies were developed using Goat Anti-Mouse IgG-Peroxidase (Cat. No. A2304) and a chemiluminescent substrate. Lanes 1. HeLa TUBB3 KO (knocked out) Cell Lysate 2. HeLa wild type Results No antibody staining can be indicated in HeLa TUBB3 KO (knocked out) Cell Lysate, as the intended target has been genetically removed lane (1) and the corresponding wild type sample demonstrate recognition of the target lane (2). Equal staining of the GAPDH protein confirm the loading control equal amount of the lysates.
Immunofluorescence SH-SY-5Y cells were fixed and permeabilized with methanol followed by methanol:acetone. Fixed cells were stained with 1:400 Monoclonal Anti-β-Tubulin III Clone: SDL.3D10 (Cat. No. T8660). The antibody was developed using Goat Anti-Mouse IgG, Cy3™ conjugate. Cells were counterstained with DAPI (blue) to stain nuclei.
Immunoblotting Cell line lysates were separated on SDS-PAGE and probed with 1:400 Monoclonal Anti-β-Tubulin III Clone: SDL.3D10 (Cat. No. T8660). The antibody was developed using Goat Anti-Mouse IgG-Peroxidase (Cat. No. A2304) and a chemiluminescent substrate. Lanes 1. U87 2. MDBK 3. RAT2

Biochem/physiol Actions: Tubulin β 3 class III (TUBB3) plays a vital role in nervous system development and axon guidance. Mutation of the gene encoding protein leads to the ocular motility disorder, congenital fibrosis of the extraocular muscle type 3 (CFEOM3), and various neurological syndromes. Altered expression of the protein affects microtubule dynamics and microtubule-kinesin interactions. Increased expression of TUBB3 is observed in tumor tissues. Decreased expression of TUBB3 can be considered as an important marker for poor prognosis of cutaneous malignant melanoma. TUBB3 is implicated in the suppression of invasive growth of tumor tissue;therefore, it is considered to be a potential target for development of antitumor drugs.

General description: Monoclonal Anti-μ Tubulin Isotype III (mouse IgG2b isotype) is derived from the SDL.3D10 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice.
General description: Tubulin β 3 class III (TUBB3) also known as β-Tubulin III, is encoded by the gene mapped to human chromosome 16q24.3. TUBB3 protein expression is restricted to neurons.
Immunogen: synthetic peptide corresponding to the C-terminal sequence of human β-tubulin isotype III coupled to BSA
Specificity: The antibody specifically recognizes an epitope located on isotype III of β-tubulin. No cross-reactivity with other tubulin isotypes is observed. The antibody can stimulate microtubule assembly when reconstituted with tubulin and either tau or MAP2.
