Purity
The purity of human, recombinant VEGF165 expressed in HEK 293 cells (Cat. No. V5765) was assessed on SDS-PAGE (1 μg/lane) followed by InstantBlue™ staining (Cat. No. ISB1L). (Representative result.)
Lane +DTT: Reduced
Lane -DTT: Non-reduced
Deglycosylation
Human, recombinant VEGF165 expressed in HEK 293 cells (Cat. No. V5765) (lanes 1-2) and VEGF165 expressed in E. coli (lanes 3-4) were either treated with N glycosidase F (lanes 1,3) or untreated (lanes 2,4) and separated on SDS-PAGE followed by InstantBlue™ staining (Cat. No. ISB1L). (Representative result.)
Lanes
1. HEK 293 VEGF165 + N glycosidase F
2. HEK 293 VEGF165 - N glycosidase F
3. E. coli VEGF165 + N glycosidase F
4. E. coli VEGF165 - N glycosidase F
Bioassay The biological activity of human, recombinant VEGF165 expressed in HEK293 cells (Cat. No. V5765) versus control VEGF165 from E. coli was measured in a cell proliferation assay using HUVEC cells. The EC50 is defined as the effective concentration that elicits a 50% increase in cell growth in a cell-based bioassay. (Representative result.)

Biochem/physiol Actions: VEGF165 belongs to the PDGF/VEGF growth factor family characterized by the presence of eight conserved cysteine residues and a cystine knot structure. VEGF is secreted by the majority of tumor cells and initiates angiogenesis by activating endothelial cells of existing blood vessels and promoting their migration.
VEGF has also been implicated in correlation with poor prognosis in breast cancer.
In addition, VEGF is released in rheumatoid arthritis in response to TNF-α, increasing endothelial permeability and stimulating angiogenesis (formation of capillaries).
General description: VEGF165 is expressed in human HEK 293 cells as a glycosylated homodimer with an apparent molecular mass of 45 kDa. Production in human HEK 293 cells offers authentic glycosylation. Glycosylation contributes to stability in cell growth media and other applications.
