Enhanced Validation-By Recombinant Expression Antibody recognition: Anti-VAMP-8 antibody produced in Rabbit (Cat. No. V7514) is confirmed by western blot in the cell sample that has been altered to overexpress the target protein-HEK-293T over expressing VAMP-8 (Lane 1) while no staining is detected in the wild type sample-HEK-293T (Lane 2). The antibody was developed with Goat Anti-Rabbit IgG, Peroxidase conjugate (Cat. No. A0545) and a chemiluminescent substrate. Antibody dilution: 1:3000.
Immunoblotting Cell lysates were separated on SDS-PAGE and blotted with Anti-VAMP8 (Cat. No. V7514). The antibody was developed with Goat anti-Rabbit IgG, Peroxidase conjugate (Cat. No. A0545) and a chemiluminescent substrate. Antibody dilution 1:3,000. Lanes 1,3. HEK293T cells transfected with human VAMP-8 2. HEK293T cells untransfected 3. Immunoprecipitation Rabbit Anti-VAMP8 (Cat. No. V7514) was used to immunoprecipitate VAMP8 from HeLa cells lysate. Detection antibody: Rabbit Anti-VAMP8 (N-terminal) (Cat. No. V7389), followed by Anti-Rabbit IgG-peroxidase (Cat. No. A0545) and a chemiluminescent substrate. Lanes 1. 1 μg antibody-IP 2. Total cell lysate 3. Negative control: flowthrough 4. Application: Anti-VAMP-8 antibody has been used in immunoblotting and immunoprecipitation.
Biochem/physiol Actions: Vesicle-associated membrane protein 8 (VAMP-8/endobrevin) is as an endosomal v-SNARE (vesicles or granules-soluble NSF attachment protein receptors) involved in fusion between early and late endosomes. It can interact with syntaxin-7, syntaxin-8, and Vti1b (Vesicle transport through interaction with t-SNAREs homolog 1B) to form an endosomal fusion complex. VAMP8 interacts with syntaxin-2 and is involved in the terminal step of cytokinesis in mammalian cells. VAMP8 is the major vesicular v-SNARE of zymogen granules from pancreatic acinar cells and has been shown to be a general vesicular SNARE for regulated exocytosis of the exocrine system. For platelet release reaction VAMP8 is the primary v-SNARE.

General description: Vesicle-associated membrane protein 8 (VAMP-8/endobrevin) is located at 2p11.2 on the human chromosome.
Immunogen: synthetic peptide corresponding to amino acids 38-56 of human VAMP8, conjugated to KLH. This sequence is highly conserved in mouse and rat VAMP8 (single amino acid substitution).
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
