Biochem/physiol Actions: | Dihydrofolate reductase (DHFR) is a folate-metabolizing enzyme that catalyzes nicotinamide adenine dinucleotide phosphate (NADPH)-dependent reduction of dihydrofolate (DHF) to tetrahydrofolate (THF), as well as folic acid to DHF. The encoded protein is implicated in regulation of intracellular folate homeostasis and it acts as a key target for cytostatic drugs. Genetic variations in the gene is associated with the development of various neurologic diseases. |

General description: | Dihydrofolate reductase converts dihydrofolate into tetrahydrofolate, a methyl group shuttle required for the de novo synthesis of purines, thymidylic acid, and certain amino acids. While the functional dihydrofolate reductase gene has been mapped to chromosome 5, multiple intronless processed pseudogenes or dihydrofolate reductase-like genes have been identified on separate chromosomes.
