| Biochem/physiol Actions: | TNKS (tankyrase) is a regulator of Wnt-signaling. It negatively regulates Axin1, thereby suppressing β-catenin degradation and causing hyperactivation of Wnt/β-catenin signaling. TNKS controls cell death and telomere length. It might also be associated with osteoarthritis. In murine lung cancer model, silencing of this gene reduces lung cancer cell growth. Application of TNKS inhibitor suppresses growth of hepatocellular carcinoma cells. Absence of TNKS activity helps in replication of human cytomegalovirus (HCMV). |

It belongs to the PARP (poly (ADP-ribose) polymerase) family of proteins and is responsible for addition of poly (ADP-ribose) (PAR) to substrate proteins. The TNKS protein contains ankyrin repeats and a sterile α-motif (SAM). |
