Biochem/physiol Actions: | The gene TNFRSF10B (TNF receptor superfamily member 10b) is a tumor suppressor gene, which upon mutation, leads to a loss of chromosome p arm, a common occurrence in head and neck tumors. It inhibits tumorigenesis via apoptosis. The cytoplasmic death domain (DD) of the protein employs fas-associated death domain (FADD) and caspases to form the death-inducing signal complex (DISC) when the receptor is trimerized. This leads to activation of caspases and cell death. |

General description: | The protein encoded by this gene is a member of the TNF-receptor superfamily, and contains an intracellular death domain. This receptor can be activated by tumor necrosis factor-related apoptosis inducing ligand (TNFSF10/TRAIL/APO-2L), and transduces an apoptosis signal. Studies with FADD-deficient mice suggested that FADD, a death domain containing adaptor protein, is required for the apoptosis mediated by this protein. Two transcript variants encoding different isoforms and one non-coding transcript have been found for this gene. (provided by RefSeq) |
