Biochem/physiol Actions: | Peroxisome proliferator-activated receptor γ coactivator (PGC)-1α is implicated in the modulation of the expression of mitochondrial oxidative phosphorylation (OXPHOS) genes and endogenous antioxidants. Variation in the gene expression leads to Huntington′s disease (HD) and type 2 diabetes in humans. PGC-1α functions as a ‘molecular switch’ in genetic pathways involved in maintaining glucose homeostasis in liver and muscle, β cell insulin secretion and mitochondrial biogenesis. Addition to this, PGC-1α has a crucial role to play in adaptive thermogenesis, skeletal muscle fiber type switching, and heart development. PGC-1α reduces or improves muscle dystrophy muscle dystrophy by stimulating various molecular pathways; therefore, increase in the concentration and activity of PGC-1 is considered to be a potential method for Duchenne muscular dystrophy (DMD) treatment. |

General description: | The protein encoded by this gene is a transcriptional coactivator that regulates the genes involved in energy metabolism. This protein interacts with PPARgamma, which permits the interaction of this protein with multiple transcription factors. This protein can interact with, and regulate the activities of, cAMP response element binding protein (CREB) and nuclear respiratory factors (NRFs). It provides a direct link between external physiological stimuli and the regulation of mitochondrial biogenesis, and is a major factor that regulates muscle fiber type determination.
