Biochem/physiol Actions: The gene DDX20 (DEAD (Asp-Glu-Ala-Asp) box polypeptide 20) encodes a RNA helicase that forms a part of the SMN (survival-of-motor-neurons) complex, which is involved in the processing and assembly of RNPs (ribonucleoproteins), such as, snRNPs (spliceosomes), snoRNPs, miRNPs, hnRNPs and transcriptosomes. It plays an essential role in early embryonic development, where it is involved in silencing of maternal transcripts and processing of RNA. DDX20 plays a part in hind brain development where it suppresses the transcription of Egr2 (early growth response 2) gene. It regulates macrophage differentiation and proliferation by silencing the expression of Ets target genes involved in Ras pathway. It also acts as a cofactor in the SUMOylation of SF-1 (steroidogenic factor 1) by PIASy (protein inhibitor of activated STAT Y). This protein facilitates the assembly of the core of SMN complex, and this activity of DDX20 is associated with spinal muscular atrophy (SMA). DDX20 acts as an oncogene that facilitates metastasis, in human breast cancer, and has potential as a therapeutic target for the same.

General description: DEAD box proteins, characterized by the conserved motif Asp-Glu-Ala-Asp (DEAD), are putative RNA helicases. They are implicated in a number of cellular processes involving alteration of RNA secondary structure such as translation initiation, nuclear and mitochondrial splicing, and ribosome and spliceosome assembly. Based on their distribution patterns, some members of this family are believed to be involved in embryogenesis, spermatogenesis, and cellular growth and division. This gene encodes a DEAD box protein, which has an ATPase activity and is a component of the survival of motor neurons (SMN) complex. This protein interacts directly with SMN, the spinal muscular atrophy gene product, and may play a catalytic role in the function of the SMN complex on RNPs. (provided by RefSeq)
