Immunofluorescence NIH-3T3 Mouse fibroblasts cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.1% Triton X-100. Fixed cells were stained with 2.5 μg/mL Anti-YB-1 (C-terminal) antibody produced in Rabbit (Cat. No. Y0396) followed by 1:500 Anti-Rabbit IgG (whole molecule)-Cy3. Picture B: cells were counterstained with DAPI (blue) to stain nuclei.
Western Blotting PC-12 cells lysate were separated on SDS-PAGE and probed with Anti-YB-1 (C-terminal) antibody produced in Rabbit (Cat. No. Y0396). The antibody was developed using 1:10,000 Anti-Rabbit IgG (whole molecule)-Peroxidase antibody produced in Goat (Cat. No. A0545). Lanes 1. 0.5 μg/mL antibody 2. 1 μg/mL antibody 3. Negative Control

Biochem/physiol Actions: Y-box-binding protein (YB-1) is shown to bind to the Y-box promoter element to regulate gene expression. It plays an important role in a wide variety of cellular functions such as transcriptional and translation regulation, DNA repair, drug resistance, and stress responses.

General description: Y-box-binding protein (YB-1) also known as nuclease-sensitive element-binding protein 1 (NSEP1) is a member of the Cold-Sock Domain (CSD) containing proteins. It is composed of three domains: the N-terminal which is thought to function as a trans-activation regulatory domain, a highly conserved nucleic acid binding domain (the CSD), and C-terminal domain, involved in protein-protein interactions.
Immunogen: synthetic peptide corresponding to amino acids 307-324 of human YB-1, conjugated to KLH. The corresponding sequence is identical in rat and mouse YB-1 proteins.
Physical form: Solution in 0.01 M phophate buffered saline, pH 7.4, containing 15 mM sodium azide.
Specificity: Anti-YB-1 (C-terminal) antibody is specific for YB-1 in rats, mice and humans. The immunizing peptide specifically inhibits the detection of the YB-1 band by immunoblotting.
