Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 10 mg/mL of non-specific GST protein (Run 1) or recombinant Influenza A virus nucleoprotein (Run 2) were analyzed by affinity binding assay using Cat. No. ZMS1020, Anti-Influenza A Nucleoprotein, clone A1 ZooMAb® Mouse Monoclonal. The binding of this antibody to recombinant Influenza A nucleoprotein displayed a KD of 2.8 x 10-6 whereas binding to non-specific GST protein displayed a KD of 2.2 x 10-3.
ELISA Enhanced Validation - Recombinant Antibody Technology 0.1 mg of recombinant Influenza A Nucleoprotein was analyzed by ELISA using various dilutions (starting at 1 mg/mL) of Cat. No. Anti-Influenza A Nucleoprotein, clone A1 ZooMAb® Mouse Monoclonal. Alkaline phosphatase-conjugated Goat Anti-Mouse secondary antibody was used for detection.

Physical form: Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Specificity: Clone A1 is a ZooMAb® mouse recombinant monoclonal antibody that detects Nucleoprotein N1 of Influenza A virus.
Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description: Nucleoprotein (UniProt: P03466; also known as Nucleocapsid protein, Protein N) is encoded by the NP gene (Gene ID: 956531) in Influenza A virus. Influenza A viruses belong to the Orthomyxoviridae family and encapsidates the negative strand viral RNA that protects it from the action of nucleases. Its envelope membrane is reported to carry hemagglutinin, neuraminidase, and the ion channel protein M2. Inside the membrane there are layers of matrix protein 1 and ribonucleoprotein (RNP) complexes that contain viral RNA and binding proteins. RNP serves as template for transcription and replication and must be localized in the host cell nucleus to start an infectious cycle. However, due to its large size, RNP cannot diffuse through the nuclear pore complex and is transported through CRM-1-dependent nuclear export machinery. Viral nucleoprotein contains two nuclear localization signals (NLS) that are responsible for active RNP import into the nucleus. It has been reported that M1 interaction with RNP hides nucleoprotein′s NLS and soon after a virion infects a new cell, M1 dissociates from the RNP under acidification of the virion driven by M2 protein. This dissociation of M1 from RNP unmasks nucleoprotein′s NLS, targeting the RNP to the nucleus. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
