Western Blotting Enhanced Validation - Recombinant Antibody Technology Myc-tagged human recombinant p53 protein was probed with Cat. No. ZMS1032, Anti-Myc tag, clone 9E10 ZooMAb® Mouse Monoclonal (1:10,000 dilution). Proteins were visualized using a Goat Anti-Mouse IgG1 secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates Myc tag (~55 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 10 mg/mL of non-specific GST protein (Run 1) or recombinant Myc-Tagged human recombinant p53 protein (Run 2) were analyzed by affinity binding assay using Cat. No. ZMS1032, Anti-Myc tag, clone 9E10 ZooMAb® Mouse Monoclonal. The binding of this antibody to Myc-Tagged p53 protein displayed a KD of 9.3 x 10-11 whereas binding to non-specific GST protein displayed a KD of 5.7 x 10-7.
ELISA Enhanced Validation - Recombinant Antibody Technology 0.1 mg of Myc-tagged human recombinant p53 protein (Blue) and GST protein (Orange) were analyzed by ELISA using three-fold dilutions (from 3000 ng/mL to 0.15 ng/mL) of Cat. No. ZMS1032, Anti-Myc tag, clone 9E10 ZooMAb® Mouse Monoclonal. Alkaline phosphatase-conjugated Goat Anti-Mouse secondary antibody was used for detection.

Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description: Identification of the target epitope is important for characterization of a monoclonal antibodies. Epitope tags are short, linear antibody recognition sequences that enable detection of tagged fusion proteins by antibodies. Over the years a wide range of anti-tag antibodies have been developed and each one of these offer unique advantages and disadvantages. Epitope tags can undergo post-translational modifications, such as phosphorylation and glycosylation in cells and cause an increase in the molecular mass of the fused protein that can result in changes in gel mobility. Epitope tags of smaller number of amino acid residues are advantageous in that they exhibit minimal side effects on structure and function of fused target protein. A Myc tag can be used in many different immunoassays and is useful when there is no antibody against the protein of interest. Adding a Myc-tag permits the detection of such proteins with an antibody against the Myc epitope. Clone 4A6 is an antibody that recognizes ten amino acid sequence EQKLISEEDL (Glu-Gln-Lys-Leu-Iso-Ser-Glu-Glu-Asp-Leu) corresponding to the C-terminal region of human c-Myc protein (UniProt: P01106). This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
