Target description: | Caspase-8 (UniProt: Q14790; also known as EC:3.4.22.61, CASP-8, Apoptotic cysteine protease, Apoptotic protease Mch-5, CAP4, FADD-homologous ICE/ced-3-like protease, FADD-like ICE, FLICE, ICE-like apoptotic protease 5, MORT1-associated ced-3 homolog, MACH) is encoded by the CASP8 (also known as MCH5) gene (Gene ID: 841) in human. Caspase-8 is a cysteine protease that plays a major role in the extrinsic apoptotic signaling pathway via death receptors. In its inactive form, it associates with FADD by interactions between their DED motifs and within the FAS-FADD-Caspase 8 complex caspase 8 undergoes self-cleavage to convert to the active form. Activated caspase 8 is subsequently released to the cytosol and it cleaves downstream effector caspases 3, 4, 6, 7, 9 and 10. Highest expression of caspase-8 is found in peripheral blood leukocytes, spleen, thymus, and liver and it is barely detectable in brain, testis, and skeletal muscle. Caspase-8 is produced with two propeptides (aa 1-216 and 375-384) that are cleaved under appropriate signals to produce a p18 (aa 217-374) and a p10 (aa 385-479) subunit. Active caspase 8 is a heterotetramer that consists of two p18 and two p10 subunits. In human, nine different isoforms of caspase-8 have been described that are produced by alternative splicing. Isoforms 5, 6, 7, and 8 lack the e catalytic site and may interfere with the pro-apoptotic activity of the complex.
