Target description: | Acylglycerol kinase, mitochondrial (UniProt: Q53H12; also known as EC:2.7.1.107, hAGK, Multiple substrate lipid kinase, HsMuLK, MuLK, Multi-substrate lipid kinase) is encoded by the AGK (also known as MULK) gene (Gene ID: 55750) in human. AGK is an inner mitochondrial membrane enzyme that is highly expressed in muscle, heart, kidney, and brain. It is a multi-substrate lipid kinase with a typical two-domain fold (DGK1 and 2) that can phosphorylate both monoacylglycerol and diacylglycerol to form lysophosphatidic acid (LPA) and phosphatidic acid (PA), respectively. It has an N-terminal 1 helix, which is complete across the membrane, and a C-terminal key region bound to the membrane. Independently of its lipid kinase activity, it also acts as a component of the TIM22 complex and plays a role in stabilizing the TIM22 complex and regulating the import and assembly of mitochondrial carrier proteins. Overexpression of AGK is reported to increase the formation and secretion of LPA, resulting in transactivation of EGFR and activation of the downstream MAPK signal transduction pathway that leads to increased cell growth. AGK has been identified as an oncogene and its overexpression of is observed in several tumor types, including prostate cancer, breast cancer, and cervical squamous cell carcinoma.
