Target description: | DNA polymerase beta (UniProt: P06746; also known as EC:2.7.7.7, DNAP, Polβ, DNA pol beta) is encoded by the POLB gene (Gene ID: 5423) in human. Pol β is a member of the X family of DNA polymerases. It plays a role in the base excision repair pathway that is essential to cleanse the genome of simple base lesions and abasic sites that arise in DNA from spontaneous base loss. Polβ is cytoplasmic under normal conditions, but it translocates to the nucleus following DNA damage where it associates with AP endonuclease 1 (Apex) to carry out DNA repair. It possesses two catalytic activities: DNA polymerase activity and dRP lyase activity. The active site for each of these activities is located on a distinct domain of the protein: 31 kDa polymerase domain and amino-terminal 8 kDa lyase domain. Polβ does not have proofreading activity; however, its accuracy in vitro results exclusively from the nucleotide selectivity of this enzyme. Aberrant Polβ overexpression results in an increased rate of spontaneous mutagenesis and a highly mutagenic tolerance phenotype and about thirty percent of all tumors are reported to harbor mutations in the POLB gene.
