The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Target description: | Calreticulin (UniProt: P27797; also known as CRP55, Calregulin, Endoplasmic reticulum resident protein 60, ERp60, HACBP, grp60) is encoded by the CALR (also known as CRTC) gene (Gene ID: 811) in human. Calreticulin is a highly conserved, monomeric, endoplasmic reticulum protein that is synthesized with a signal peptide (aa 1-17), which is subsequently cleaved off to generate the mature form (aa 18-417). It serves as a calcium-binding chaperone that promotes folding, oligomeric assembly, and quality control in the endoplasmic reticulum via the calreticulin/calnexin cycle. It consists of three distinct domains: An N-terminal lectin-like globular domain that is required for its interaction with integrins and it contains a steroid receptor-like DNA-binding site that is involved in chaperone functions; a second domain is a central proline-rich domain that forms an elongated arm-like structure and has high-affinity and low-capacity for calcium-binding and participates in its chaperone activity. The third domain is a highly acidic, localized in the C-terminal region, displays low-affinity and high-capacity for Ca2+ binding and is involved in calcium -buffering functions of calreticulin. It also has a KDEL motif at the C-terminus that is responsible for calreticulin retrieval from the Golgi apparatus to the endoplasmic reticulum. Cancer cells undergoing immunogenic cell death are reported to expose calreticulin on their surface, which promotes the uptake of dead cells by phagocytes. Mutations leading to loss-of-function of CALR gene promote oncogenesis by impairing homeostasis in healthy cells and hamper natural and therapy-driven immunosurveillance.
