Target description: | RNA-binding protein Nova-2 (UniProt: Q9UNW9; also known as Astrocytic NOVA1-like RNA-binding protein, Neuro-oncological ventral antigen 2) is encoded by the NOVA2 (also known as ANOVA, NOVA3) gene (Gene ID: 4858) in human. The neuron specific RNA-binding proteins NOVA1 and NOVA2 are highly homologous alternative splicing regulators. Both NOVA1 and NOVA2 have three K homology (KH) domains. They are described as autoantigens in the human autoimmune neurological disease known as paraneoplastic opsoclonus-myoclonus ataxia (POMA) that is characterized by loss of motor control in limbs and eyes. NOVA proteins regulate alternative splicing in neurons by binding pre-mRNA in a sequence-specific manner to activate exon inclusion or exclusion. NOVA2 binds RNA with high affinity and with sequence specificity that differs from NOVA1. Its expression is restricted o astrocytes. In close association with NOVA 1, it performs unique biological functions in different brain areas and cell types. NOVA2 binding to an exonic 5′-YCAY-3′ cluster changes the protein complexes assembled on pre-mRNA, blocking U1 snRNP binding and exon inclusion, whereas binding to an intronic 5′-YCAY-3′ cluster enhances spliceosome assembly and exon inclusion. It is reported to regulate alternative splicing events of several axon guidance related genes during cortical development. It controls RNA splicing in both inhibitory and excitatory neurons.
