Western Blotting Enhanced Validation - Recombinant Antibody Technology Lysates from HeLa (Lane 1), SH-SY5Y (Lane 2), COS-1 (Lane 3) cells probed with Cat. No. ZRB1856, Anti-14-3-3 β/α, clone 1D15 ZooMAb® Rabbit Monoclonal (1:10,000 dilution). Proteins were visualized using a Rabbit Anti-Donkey IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates 14-3-3 β/α protein (~28 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of 14-3-3 β/α peptide (Run 1) or non-specific control peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1856, Anti-14-3-3 β/α, clone 1D15 ZooMAb® Rabbit Monoclonal. The binding of this antibody to 14-3-3 β/α peptide displayed a KD value of 2.3 x 10-6, whereas its binding to non-specific control peptide displayed a KD of 2.0 x 10-3.
ELISA Enhanced Validation - Recombinant Antibody Technology 14-3-3 β/α protein was analyzed using two-fold serial dilutions of Cat. No. ZRB1856, Anti-14-3-3 β/α, clone 1D15 ZooMAb® Rabbit Monoclonal. Plate was coated with 100 mL of recombinant 14-3-3 β/α protein (1 mg/mL). Protein was detected using alkaline phosphatase conjugated Goat Anti-Rabbit IgG (H&L) and pNPP substrate.

Target description: 14-3-3 protein β/a (UniProt: P31946; also known as Protein 1054, Protein kinase C inhibitor protein 1, KCIP-1) is encoded by the YWHAB gene (Gene ID: 7529) in human. Mammals express seven distinct 14-3-3 isoforms (gamma, epsilon, beta, zeta, sigma, theta, tau) that form multiple homo- and heterodimers, thus rendering 14-3-3 as a key multifunctional regulatory molecule. 14-3-3 protein beta/alpha serves as an adapter protein that has been implicated in the regulation of a large spectrum of both general and specialized signaling pathways. It binds to many partners, usually by recognition of a phosphoserine or phosphothreonine motif and its binding generally results in the modulation of the activity of the binding partner. The alpha, brain-specific form, differs from the beta form in its ability to be phosphorylated on serine 60 by protein kinase C delta in a sphingosine-dependent fashion. 14-3-3 β/a is shown to block the nuclear translocation of the phosphorylated form of SR protein-specific kinase 2 (SRPK2) and antagonize its stimulatory effect on cyclin D1 expression that results in blockage of SRPK2 elicited neuronal apoptosis. It also reported to serve as a negative regulator of osteogenesis.
