General description: Polymer-based particles for reversed-phase (RP) HPLC generally have two benefits over silica-based particles: They are more tolerant of high and low pH operating conditions and they do not have the silanol interactions that cause peak tailing of some basic analytes. Also, some silicas have an intrinsic metal content that may cause chelation problems with sensitive biomolecules, leading to poor recovery. Polymeric TSKgel columns come in C18 and phenyl functional groups and a range of particle porosities. Fast kinetics and quantitative protein recovery at submicrogram loading are characteristic of nonporous resin-based TSKgel Octadecyl-NPR columns.

General description: TSKgel Phenyl-5PW RP columns are filled with 10 micron porous polymethacrylate particles, the surface of which is covered with a high density of phenyl groups that causes proteins to be retained in a low salt buffer mobile phase and requiring a solvent gradient for their elution, as is the protocol in reversed phase chromatography. In contrast, TSKgel Phenyl-5PW has such a low density of phenyl groups on the surface that proteins can only be retained at high salt concentration and eluted by a decreasing salt gradient, which, of course, is the protocol used in hydrophobic interaction chromatography. TSKgel Phenyl-5PW RP columns can withstand rigorous cleaning with either acid or base. The 1000 A pore size accommodates globular protein samples up to 1,000,000 Da.
